The Juju knife represents a Leopard Person’s skill and ties to magic. Without it, Sunny is unable to conjure more complex jujus, such as summoning music to help her cross the bridge to Leopard Knocks or sending away Ekwensu at the end of the novel. Essentially, the Juju knife is a key to much of the magic that a Leopard person has. This is also why the knife chooses the owner, because the link between them needs to be unbreakable.
